Protocols
This page tries to summarize the protocols we use in our work.
pGLO experiment
For the BIO-RAD pGLO experiment, there are three kinds of plates used:
- 16 plates with LB agar only
- 16 plates with LB agar and ampicillin
- 8 plates with LB agar, ambicillin and arabinose
There, they are using small petri dishes, which take about 12ml of agar.
For the plates with LB agar only, you use about 16 x 12ml = 192ml ≈ 200ml of LB agar.
For the plates with LB agar and ampicillin, you use about:
- 16 x 12ml = 192ml ≈ 200ml of LB agar
- 2ml of their ampicillin solution (that is, 10μl/ml of ampicillin)
For the plates with LB agar, ampicillin and arabinose, you use about:
- 8 x 12ml = 96ml ≈ 100ml of LB agar
- 1ml of their ampicillin solution (that is, 10μl/ml of ampicillin)
- 3ml of their arabinose solution (that is, 30μl/ml of arabinose)
